What it is
Melanotan 2 (MT-2) is a synthetic peptide that mimics α-melanocyte-stimulating hormone (α-MSH). It is best known for stimulating melanin production and skin pigmentation, with secondary research interest in sexual function and appetite control. Effects are downstream of broad melanocortin receptor activation (MC1, MC3, MC4, MC5).
Mechanism of action
Non-selective melanocortin receptor agonist. MC1 activation drives melanin synthesis; MC4 activation underlies sexual arousal and appetite suppression; MC3/5 contribute to lipid and immune effects.
Half-life: ~30 minutes to a few hours, with prolonged tissue effects on pigmentation.
Evidence summary
Pilot Phase 1 work (1996) characterised the cyclic peptide pharmacology. Subsequent trials in psychogenic ED (1998), preformulation work, and recent appetite/dermatology research extend the profile, though MT-2 is unapproved.
Typical dosing protocols
- Loading: 250–500 mcg subcutaneous daily until pigmentation onset
- Maintenance: 250–500 mcg, 1–3× weekly
- Apply standard sun-exposure for activation

- Nausea, flushing, appetite suppression early in cycle
- Spontaneous erections / yawning common (MC4 effects)
- Risk of new or darkening moles — dermatology monitoring advised
